ABSTRACT

Objective: Headspace-solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) was used to analyze the characteristic volatile compounds from 18 heat-clearing herbs. Method: HS-SPME was used to extract the volatile components from different heat-clearing herbs,and the chemical compositions were analyzed by GC-MS. The GC was performed at the injector temperature of 250℃,the high-purity helium was used as the carrier gas at a constant column flow of 1.0 mL·min-1 with no split sampling. The solvent delay time was 3 min. Mass spectrum conditions included electron bombardment ion source,electron bombardment energy of 70 eV,ion source temperature of 230℃,quadrupoles temperature of 150℃,and scanning range of m/z 29-370.The volatile components were confirmed according to NIST2013 database,and the relative content percentage of each component was calculated by peak area normalization method. PCA was used to detect clustering and discuss the characteristics of volatile compounds of 18 heat-clearing herbs. Result: A total of 328 volatile compounds were detected in 18 heat-clearing herbs,including alkanes,unsaturated hydrocarbon,alcohols,aldehyde,ketones,esters,furan and other unknown compounds. The number and relative content of volatile components in different heat-clearing herbs varied dramatically. Several important characteristic volatile compounds,such as l-camphor,torreyol,caryophyllene oxide,globulol and 2,4-cycloheptadien-1-one,2,6,6-trimethyl,were detected in the 18 heat-clearing herbs by principal component analysis. Conclusion: Heat-clearing herbs contains rich volatile components,which are the main contributors to the formation of unique aroma of herbs,as well as the main active substances.

ABSTRACT

OBJECTIVE: To prepare amphotericin B vaginal nanosuspension foam and study the physicochemical properties and in vivo pharmacodynamics. METHODS: Using the single factor method, the concentration of egg lecithin and the technical parameters of foaming agent in formulation were gradually optimized. Then, the physicochemical properties, including drug particle size, appearance of foam and formed capability, were studied. The therapeutic effect on vaginal inflammatory infection was investigated with the mouse model of Candida albicans vaginitis. RESULTS: The optimal formulation of amphotericin B nanosuspension foam included 5 mg•mL-1 amphotericin B, 60 mg•mL-1 egg lecithin and 30 mg•mL-1 APG0810. The drug particle size, range of micro-foam diameter and foam half-life were about 240 nm, 200-400 μm and 43.5 min, respectively. It was found that amphotericin B nanosuspension vaginal foam had significant therapeutic effect on mouse vaginal inflammation infection. CONCLUSION: The amphotericin B nanosuspension foam has good physicochemical properties and excellent pharmacological activity in vivo, which shows potential use for the therapy of vulvovaginal candidiasis.

ABSTRACT

The clinical thinking modes of "integrative disease and Syndrome" and "integrative macroscopical Syndrome-differentiation and microscopical Syndrome-differentiation" formed in the past half century were merely the primary train of thought and methodology of integrative traditional Chinese and Western medicine. In order to develop them to a higher level, stress should be put on the unity and holism of disordered organism and unity of function, structure and metabolism. It has long been ignored that a parallel and correlated relationship exists between the TCM Syndrome and pathological process of Western medicine, which is a common, set-formed and regular pathophysiological changes with certain development phase, presented in various diseases. Both have common features as that the same pathological process can be seen in different diseases and several pathological progresses can present in one disease, the same Syndrome also can be seen in different diseases and several Syndromes can present in a disease. Moreover, they have the compatible connotation and denotation. The author deduced that pathophysiological basis of main Syndrome is indeed the basic pathological process. Essence of Syndrome is the related pathological processes including abnormal changes in function, metabolism and morphological structure.
